How to test for Fecal Coliform

According to the State of North Carolina, Fecal Coliform may not exceed a geometric mean of 200/100 ml from at least 5 samples during a 30 day period.

Furthermore Fecal Coliform is not to exceed 400/100 ml in more than 20 percent of the samples tested during those 30 days.

However any presence of Fecal Coliform in water samples indicates that the water has been contaminated
Fecal Coliform Methods

This is the method using the P/A (presence/absence) broth. It is simple to use and all of the components come prepackaged and pre-measured.
• Collect 100 mL of sample in a sterile container
• Fill the sample to the fill-to line of a sample bottle
• Add one P/A Broth ampule to the 100 mL sample
• At 35 +- 0.5 degrees Celsius incubate the sample for 24-48 hours
• Observe the reaction after 24 hours of being incubated
• See the interpreting and reporting results on table 1
• Confirm a positive sample by vaccinating the appropriate source directly from the positive P/A broth cultures
• See the confirmation table 2
• Dispose of all completed tests in their designated waste container

Reactions using P/A Broth

Possible Positives for total coliform bacteria
• If the color changes from reddish purple to yellow or yellow brown after 24 hours or after 48 hours

Negative for total coliform bacteria
• If no color change and the color is still reddish pruple even after 48 hours

Reactions using P/A Broth with MUG

Positive for E.Coli
• If under long-wave UV light the sample fluoresces

Negative for E. Coli
• If under long-wave UV light the sample does not fluoresce
